Stratum Spinosum

Transmission electron microscopy (TEM) of the stratum spinosum in thin skin.

The stratum spinosum comprises several layers of keratinocytes in the middle of the epidermis. The keratinocytes are interconnected by “spinous” processes joined by desmosomes, which makes the skin strong and flexible.

  • (red/yellow/cyan/blue) - responsible for making keratin
    • Keratin Filaments (black) - intermediate filaments
    • Desmosomes (, ,)- spot-like junctional complexes for cell-to-cell adhesion
      • Attachment Plaques (purple) - dense protein plaque inside the plasma membrane connected to keratin filaments
        • Desmoplakin and plakoglobin - anchoring proteins that bind the cytoplasmic domains of cadherins to intermediate filaments
      • Cadherins (yellow) - transmembrane linker proteins that span the extracellular space between adjacent cell membranes
        • Desmoglein and desmocollin
  • Langerhan Cells (,; green) - tissue-resident macrophages that are functionally more similar to antigen-presenting dendritic cells
    • Irregular nuclei with grooves
    • No desmosomes with keratinocytes
